- Recognizes human albumin
- Albumin most abundant protein in blood plasma
- Produced by well-differentiated hepatocytes
- Plays role in maintaining osmotic pressure
- Decreased levels of serum albumin may be sign of kidney or liver diseases and diabetes
Process:
- Antibody specific for Albumin pre-coated onto microwells
- Albumin protein in samples captured by the coated antibody after incubation
- After extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for Albumin is added to detect captured Albumin
- For signal development, followed by Tetramethyl-benzidine (TMB) reagent
- Solution containing sulfuric acid used to stop color development and color intensity
- Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
